Recovery of DNA from bone without demineralization

Abstract This study assessed the performance of five different DNA extraction methods for recovery from bone: ChargeSwitch® gDNA Plant Kit, IQ™ System DNeasy® Blood & Tissue PrepFiler® BTA Forensic Extraction Kit and phenol-chloroform-isoamyl alcohol. was extracted pig rib femur bones that fresh, had undergone surface decomposition three months, one year. Extracted analyzed using real-time PCR amplification an in-house multiplex quality quantity presence inhibitors. The phenol-chloroform-based method consistently yielded highest amounts IQ lowest; however, all produced relatively high yields both samples could be amplified without any detected inhibition. data demonstrate with reasonable bone tested can isolate successfully analyzed. effective use internal controls is also demonstrated.
